TNF—α启动子基因多态性与子痫前期的相关分析(1)

     【摘要】 目的:研究腫瘤坏死因子α(TNF-α)启动子基因多态性与子痫前期的关系。方法:选取本院2014年9月-2017年9月期间收治的子痫前期患者156例作为研究组,另选取同期来本院产检无并发症的正常妊娠孕妇150例作为对照组,对两组研究对象进行TNF-α启动子基因检测,分析TNF-α启动子基因多态性与子痫前期的关系。结果:重度子痫前期患者血清TNF-α表达水平为(3.96±0.78)pg/mL

    明显高于轻度子痫前期的(3.22±0.71)pg/mL;轻度子痫前期患者血清TNF-α表达水平明显高于对照组的(2.43±0.64)pg/mL,差异均有统计学意义(P<0.05)。研究组TNF-α-238启动子基因型分布情况与对照组对比,差异均无统计学意(P>0.05);研究组TNF-α-308启动子GA和AA基因型分布率均明显高于对照组,GG基因型分布率明显低于照组,差异均有统计学意义(P<0.05)。GG、GA+AA基因型在不同程度子痫前期患者中对比,差异均无统计学意义(P>0.05)。GA+AA基因型患者血清TNF-α表达水平为(4.31±0.46)pg/mL,明显高于GG基因型的(3.25±0.63)pg/mL,差异有统计学意义(P<0.05)。结论:TNF-α-308启动子基因多态性可影响细胞因子的表达水平,可能参与了子痫前期的发病过程。

    【Abstract】 Objective:To study the correlation of tumor necrosis factor α (TNF-α) promoter gene polymorphism and pre-eclampsia. Method:A total of 156 patients with pre-eclampsia treated from September 2014 to September 2017 in our hospital were selected as the study group. In addition,150 normal pregnant women without complications at the same period were selected as the control group.The TNF-α promoter gene was detected in two groups,and the relationship between TNF-α promoter polymorphism and pre-eclampsia was analyzed.

    Result:The level of serum TNF-α expression in patients with severe pre-eclampsia was (3.96±0.78)pg/mL,which was significantly higher than (3.22±0.71)pg/mL in mild pre-eclampsia;the level of serum TNF-α expression in the patients with mild pre-eclampsia was significantly higher than (2.43±0.64)pg/mL in the control group,the differences were statistically significant(P<0.05).The TNF-α-238 promoter gene distribution rate in the study group was not significantly different from that in the control group (P>0.05).The TNF-α-308 promoter GG and AA genotypes distribution rates in the study group were significantly higher than that in the control group;the GA genotype distribution rate was significantly lower than that in the control group,the differences were statistically significant (P<0.05).There were no significant differences in GG,GA+AA genotype distribution rates in patients with different degree of pre-eclampsia(P>0.05).The level of serum TNF-α expression in patients with GA+AA genotypes were (4.31±0.46)pg/mL,which was significantly higher than (3.25±0.63)pg/mL in patients with GG genotype,the difference was statistically significant(P<0.05).Conclusion:The TNFα-308 promoter gene polymorphism will affect the level of cell factor expression and be involved into the genesis process of pre-eclampsia.
